摘要

帕金森病(Parkinson's disease, PD)是世界上第二常见的神经退行性疾病,发病率呈逐年上升趋势, 其病理特征是α-突触核蛋白(α-synuclein, α-Syn)的异常聚集。内质网在维持蛋白质稳态中起重要作用, 许多研究表明该细胞器参与了PD及其他神经退行性疾病的进展。病理情况下, 包括α-Syn在内的各种未折叠或异常折叠蛋白可能引起内质网应激(endoplasmic reticulum stress, ERS), 干扰细胞正常功能, 然而α-Syn诱发ERS的分子机制尚未完全明确。本文旨在概述PD进程与病理性α-Syn和ERS的相关性,以及目前的治疗研究进展,为PD的诊断和治疗提供新思路。

Abstract

Parkinson's disease (PD) is the second most common neurodegenerative disease worldwide, with an increasing incidence over the years. Its pathology is characterized by the abnormal aggregation of α-synuclein (α-Syn). The endoplasmic reticulum plays a critical role in maintaining protein homeostasis, and numerous studies have shown its involvement in the progression of PD and other neurodegenerative diseases. Under pathological conditions, various unfolded or misfolded proteins, including α-Syn, may cause endoplasmic reticulum stress (ERS) and interfere with normal cellular function. However, the molecular mechanism of ERS induced by α-Syn has not been fully elucidated. The article aims to summarize the correlation of α-Syn and ERS with the progression of PD, as well as the current progress in therapeutic research, thus providing new insights for the diagnosis and treatment of PD.
