摘要

瞬时受体电位香草酸 3(transientreceptorpotentialvanilloid3,TRPV3)是 Ca2+ 通透的非选择阳离子通道,可被>33℃温度激活。TRPV3在皮肤屏障修复、瘙痒和炎症等疾病的发展过程中扮演重要的角色,其点突变导致以过度角化、瘙痒和脱发为特征的罕见人类遗传病—Olmsted 综合征。研究表明,TRPV3是瘙痒和皮肤疾病的潜在治疗靶点,特异性抑制 TRPV3 可能具有应用前景。本文就 TRPV3 结构与功能及其病理特别是皮肤疾病相关病理的最新研究进展作一综述。

Abstract

Transient receptor potential vanilloid 3 (TRPV3) is a warm-temperature sensitive calcium-permeable nonselective cation channel that is activated by innocuous heat above 33℃. TRPV3 is involved in skin barrier repair, pruritus and inflammation, and its genetic mutations lead to a rare genetic disease called Olmsted syndrome characterized by bilateral mutilating transgredient palmoplantar keratoderma (PPK), periorificial keratotic plaques and chronic pruritus. These findings indicate that TRPV3 is likely to serve as a drug target and selective inhibition of TRPV3 may hold therapeutic potential for treatment of pruritus and skin-related diseases. This review outlines recent progress in the structure and function of TRPV3,as well as its pathological roles in skin diseases.
